Use, storage and stability
Mouse RSPO1-his protein has been successfully used in various types of organoids like colon, ileum and duodenum organoids. The working concentration of the product in organoid cultures should be determined experimentally. As a starting concentration 10-20 nM can be used. Depending on the type of organoid 0.2 nM WNT surrogate-Fc fusion protein (catalog U-N001), or similar, can be added.

The product should be stored at -80°C. After thawing it should be stored in appropriate small aliquots at -20°C or -80°C (stable for at least 6 months) or at 4°C for 3 weeks.

Production platform:
This product has been produced in HEK293 cells using the rPExTM recombinant protein production platform.

Background Description
RSPO1 is a secreted activator protein with two cysteine-rich, furin-like domains and one thrombospondin type 1 domain. The encoded protein is a ligand for leucine-rich repeat-containing G-protein coupled receptors (LGR proteins) and positively regulates the Wnt/β-catenin signaling pathway. In mice, the protein induces the rapid onset of crypt cell proliferation and increases intestinal epithelial healing, providing a protective effect against chemotherapy-induced adverse effects. Although R-spondins are unable to initiate Wnt signaling, they can potently enhance responses to low-dose Wnt proteins.
