mRSPO1-Fc Fusion Protein | ||
Use, storage and stability Mouse RSPO1-Fc fusion protein has been successfully used in various types of organoids like colon, ileum and duodenum organoids. The working concentration of the product in organoidcultures should be determined experimentally. As a starting concentration 10 – 20 nM can be used. Depending on the type of organoid 0.2 nM WNT surrogate-Fc fusion protein (catalog U-N001), or similar, can be added. The product should be stored at -80°C. After thawing it should be stored in appropriate small aliquots at -20°C or -80°C (stable for at least 6 months) or at 4°C for 3 weeks. Background | Description RSPO1 is a secreted activator protein with two cysteine-rich, furin-like domains and one thrombospondin type 1 domain. The encoded protein is a ligand for leucine-rich repeat-containing G-protein coupled receptors (LGR proteins) and positively regulates the Wnt/β-catenin signaling pathway. In mice, the protein induces the rapid onset of crypt cell proliferation and increases intestinal epithelial healing, providing a protective effect against chemotherapy-induced adverse effects. Although R-spondins are unable to initiate Wnt signaling, they can potently enhance responses to low-dose Wnt proteins. |
