Background mRNA surveillance is the quality control mechanism to detect and eliminate abnormal mRNAs in eukaryotes. Nonsense-mediated mRNA decay (NMD) is one of the mRNA surveillance systems under which mRNAs containing pre-termination codons (PTCs) are selectively degraded. UPF3B, a core component of NMD machinery, is involved in various processes such as mRNA nuclear export, NMD and mRNA translation. In nucleus, UPF3B associates with exon junction complex (EJC) on mRNA and travels to cytoplasm. In cytoplasm, UPF3B bound EJC on mRNA interacts with its homologues UPF1 and UPF2, and promotes NMD by enhancing ATP-dependent helicase activity of UPF1 together with UPF2.
