| Background | Reelinは、388 kDaからなる細胞外マトリックスの糖タンパク質であり、セリンプロテアーゼ活性を持つ。ReelinはApoER2/VLDLRの２つのReelin受容体と結合したのち，細胞内タンパク質であるDab1をリン酸化する。 Reelinの構造は、シグナル配列、SP配列に続いてReelin特異的なH配列が存在し、その後ろに300～350残基のアミノ酸配列が8回繰り返されるReelin repeatsで構成されている。リピート2と3の間のN-t cleavage、およびリピート6と7の間のC-t cleavageによって切断を受ける。本モノクローナル抗体は、R2切断部位をまたぐペプチドを免疫し、N-t cleavageを受けたReelinを認識しない抗体である。N-t cleavageによりReelinは不活性化されるため、本モノクローナル抗体は、活性型Reelinを特異的に認識するモノクローナル抗体である。 Reelinは、脳において神経細胞の移動や発達過程の脳の位置決定などに機能し、樹状突起、樹状突起棘の発達を促進する。Reelinの異常は脳構造の異常を導き、統合失調症や自閉症など神経変性疾患との関連も示唆されている。 |
