Background Human TLR3 consists of an extracellular domain containing 23 leucine rich repeats and N- and C-terminal flanking regions, the transmembrane domain, and the intracellular Toll-IL-1 receptor homology domain. It possesses 15 putative carbohydrate-binding motifs in the extracellular domain. Human TLR3 is expressed in myeloid DCs, macrophages, fibroblasts and a variety of epithelial cells. TLR3 localizes on endosomes in myeloid DCs and both on the cell surface and endosomes in fibroblasts, macrophages and some of epithelial cell lines. TLR3 recognizes double-stranded RNA including poly(I:C), induces the activation of NF-κB, AP-1 and IRF3 through the TICAM-1-dependent pathway, resulting in the production of IFN-γ and proinflammatory cytokines. TLR3.7 inhibits poly(I:C)-induced IFN-γ production by human fibroblasts naturally expressing TLR3 on their surface.
