Background The CRISPR/Cas9 system is an innovative tool for genome editing. In 2015, a new Cas9 gene was cloned from Staphylococcus aureus. The SaCas9 (Staphylococcus aureus Cas9) gene is 1 kb shorter than the SpCas9 (Streptococcus pyogenes Cas9) gene, which was originally used for genome editing (Ran FA, et al., 2015, Nature). The protospacer adjacent motif (PAM) sequence of SaCas9, required for DNA cleavage, is longer than that of SpCas9. These molecular characteristics of SaCas9 are expected to enable more efficient genome editing.
